چکیده

We have developed a target organism which permits quantitative bactericidal assays. The organism is an Escherichia coli mutant which cannot grow at the temperature of the assay (37 degrees C), but retains full colony-forming potential for subsequent quantitation at 25 degrees C. We show that quantitative data on the bactericidal capacity of polymorphonuclear leukocytes and alveolar macrophages can be obtained when this mutant is used as a target. The procedure used to generate the strain is described in detail and should be applicable to many bacterial species. Characterization of the properties of the mutant indicates that it has a strong potential for use in other in vivo and in vitro investigations of host responses to microbial invasion.

Quantifying and comparing constitutive immunity across avian species.

Studies that blend a comparative approach to immunology with an appreciation for physiological ecology are defining an important new field in biology--ecological immunology. However, a panel of assays that permits a comparative approach to immunology is not yet available. In this paper, we describe several assays of innate immunity that do not require species-specific reagents and therefore ide...

Bactericidal and Fungicidal Action of Organic Mercurials

It has been stated recently (Hunter, 1943) that the ideal antiseptic should possess high germicidal and inhibitory properties over the widest possible range of the commoner pathogenic organisms, a low tissue toxicity, efficiency in the presence of organic matter, power to penetrate, stability, and moderate cost. In this country the commoner pathogenic organisms include a great number of mycotic...

Toxicity of pesticides to plants and non-target organism: A comprehensive review

Abstract Pesticides are agrochemical used to increase agronomic production and higher economic profit. However, these chemical pesticides could be persistent in soil and water, collective in silt or bio-aggregates in biota relying upon their dissolvability, prompting various kinds of natural contamination. These pesticides have also shown detrimental effects such as phytotoxicity, genotox...

Discovery and Analysis of Natural-Product Compounds Inhibiting Protein Synthesis in Pseudomonas aeruginosa.

Bacterial protein synthesis is the target for numerous natural and synthetic antibacterial agents. We have developed a poly(U) mRNA-directed aminoacylation/translation (A/T) protein synthesis system composed of phenylalanyl-tRNA synthetases (PheRS), ribosomes, and ribosomal factors from Pseudomonas aeruginosa This system has been used for high-throughput screening of a natural-compound library....

Rapid SARS-CoV-2 RT-LAMP assay setup using gene construct design approach

Background and Objective: The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is still an ongoing challenge in health system worldwide. Molecular detection methods including reverse transcription loop-mediated isothermal amplification (RT-LAMP) can help cutting off the transmission chain of the virus. In this study, prompt setting up of the RT-LAMP assays were investigated using ge...
